Biological Background: IL2 Function and Localization
- Interleukin-2 (IL2), also known as T-cell growth factor (TCGF), is a cytokine primarily produced by activated CD4-positive helper T-cells and, to a lesser extent, by activated CD8-positive T-cells and natural killer (NK) cells. It plays a central role in the immune response and tolerance.
Related references: PMID:14614860, PMID:9814585 - IL2 exerts its effects by binding to a receptor complex composed of either the high-affinity trimeric IL-2R (IL2RA, IL2RB, IL2RG) or the low-affinity dimeric IL-2R (IL2RB and IL2RG). This interaction induces oligomerization and conformational changes, leading to activation of JAK1 and JAK3 kinases and subsequent phosphorylation of STAT5.
Related references: PMID:14614860, PMID:27018889 - Downstream signaling through the IL2 receptor activates multiple pathways, including the STAT, phosphoinositide-3-kinase (PI3K), and mitogen-activated protein kinase (MAPK) pathways, which mediate its potent growth factor activity.
- As a T-cell growth factor, IL2 promotes strong proliferation of activated B-cells and immunoglobulin production, and enhances the cytolytic activity of NK cells.
- IL2 is critical for the differentiation and homeostasis of effector T-cell subsets, including Th1, Th2, Th17, and memory CD8-positive T-cells.
Related references: PMID:9814585 - This cytokine is essential for the survival and proliferation of regulatory T-cells (Tregs), which are required for the maintenance of immune tolerance.
Related references: PMID:14614860 - The protein is secreted and primarily produced by immune cells such as dendritic cells, whereas macrophages do not produce IL2 upon bacterial stimulation.
- IL2 is a glycoprotein with a molecular weight of approximately 19.4 kDa (precursor) and belongs to the IL-2 family of cytokines.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to amino acids 21–169 of mouse IL2, encompassing the mature secreted form. This design suggests the antibody likely recognizes the native and denatured form of IL2, supporting its use in WB, IF-P, and ELISA applications.
- For Western blot, consider using reduced and non-reduced samples to assess potential disulfide bond-dependent epitopes.
- In immunofluorescence, adequate permeabilization may be required to detect the secreted cytokine; however, intracellular staining of producing cells can be optimized.
- When using the antibody in ELISA, validate pairing with a suitable detection antibody for sandwich immunoassays.
