Biological Background: IL2 Function and Localization
- Interleukin-2 (IL-2), also known as T-cell growth factor (TCGF) and lymphokine, is a critical immunoregulatory cytokine.
- Produced predominantly by activated CD4+ helper T-cells, and to a lesser extent by CD8+ T-cells and natural killer (NK) cells, playing a pivotal role in immune response and tolerance (PubMed:6438535). Related references:
- PMID:6438535
- Binds to a heterotrimeric high-affinity IL-2 receptor (IL-2Rα/CD25, IL-2Rβ/CD122, γc/CD132) or a dimeric intermediate-affinity receptor (IL-2Rβ, γc) (PubMed:16293754, PubMed:16477002). Related references:
- PMID:16293754
- PMID:16477002
- Receptor engagement activates JAK1/JAK3 kinases, leading to phosphorylation and nuclear translocation of STAT5, and stimulates the PI3K/AKT and MAPK signaling cascades (PubMed:7973659, PubMed:8580378, PubMed:25142963). Related references:
- PMID:7973659
- PMID:8580378
- PMID:25142963
- Functions as a potent T-cell growth factor and enhances the cytolytic activity of NK cells (PubMed:6608729). Related references:
- PMID:6608729
- Promotes strong proliferation of activated B-cells and subsequent immunoglobulin production (PubMed:6438535). Related references:
- PMID:6438535
- Controls the survival and proliferation of regulatory T-cells (Tregs), which are essential for the maintenance of immune tolerance.
- Participates in the differentiation and homeostasis of effector T-cell subsets, including Th1, Th2, Th17, and memory CD8+ T-cells.
- The protein is secreted as a glycoprotein and contains an N-terminal signal peptide and intrachain disulfide bonds (UniProt: P60568).
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein containing the full-length mature human IL-2 sequence (amino acids 1–153). This design covers the complete extracellular domain and may recognize both glycosylated and unglycosylated protein forms.
- For Western blotting (WB), a band around 18 kDa is expected; however, glycosylation and sample preparation can cause apparent size shifts. Include appropriate positive controls and molecular weight markers.
- IL-2 is a secreted cytokine; for flow cytometry (FC), intracellular staining with a protein transport inhibitor (e.g., brefeldin A or monensin) is recommended to trap the protein inside activated cells.
- This antibody could be evaluated as a capture or detection reagent in sandwich ELISA formats when paired with a validated antibody targeting a non-overlapping epitope. Titration and cross-reactivity testing are advised.
