Biological Background: IL1β Function and Localization
- Interleukin-1 beta (IL1β) is a potent pro-inflammatory cytokine belonging to the IL-1 family. It is produced as an inactive precursor (pro-IL1β) and cleaved to its mature active form by caspase-1.
- IL1β mediates pleiotropic effects including induction of prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production.
- It promotes Th17 differentiation of T-cells and synergizes with IL12 to induce IFNG synthesis from T-helper 1 (Th1) cells.
- IL1β plays a role in angiogenesis by inducing VEGF production synergistically with TNF and IL6.
- Involved in transduction of inflammation downstream of pyroptosis: its mature form is specifically released through the gasdermin-D (GSDMD) pore.
- Subcellular localization includes cytoplasm, cytosol, lysosome, and secreted extracellularly via exosomes.
- Expressed in activated macrophages at the protein level.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 118-269 of mouse IL1β, which covers the mature secreted form. This design is expected to recognize the active cytokine in various species.
- For Western blot (WB), consider using reducing conditions and appropriate positive controls such as LPS-stimulated macrophage lysates; the predicted molecular weight is approximately 31 kDa, but post-translational modifications may affect migration.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization may be necessary. Validate in tissue samples known to express IL1β.
- ELISA applications may require pairing with a detection antibody and optimization of capture/detection conditions.
