Biological Background: IL1β Function and Localization

  • Interleukin-1 beta (IL1β) is a potent pro-inflammatory cytokine belonging to the interleukin-1 family, encoded by the Il1b gene in mouse (Gene ID: 16176, UniProt P10749). The protein is synthesized as a 31 kDa precursor that is cleaved to generate a mature secreted form.
  • IL1β was initially identified as the major endogenous pyrogen, capable of inducing fever. It stimulates prostaglandin synthesis, promotes neutrophil influx and activation, and regulates both T-cell and B-cell functions, driving adaptive immune responses.
  • IL1β plays a central role in the inflammatory cascade: it synergizes with IL12 to induce interferon-gamma (IFNG) production from Th1 cells, and promotes Th17 differentiation. It also facilitates angiogenesis by inducing VEGF production in combination with TNF and IL6.
  • The mature form of IL1β is released extracellularly via a non-classical secretion pathway. Upon inflammasome activation and pyroptosis, mature IL1β passively exits through gasdermin-D (GSDMD) pores in the plasma membrane, linking innate immune sensing to inflammation.
  • Subcellularly, IL1β is found in the cytosol, lysosomes, and is actively secreted. It is also associated with extracellular exosomes, indicating multiple secretion routes (UniProt: SUBCELLULAR LOCATION).
  • Expression of IL1β is tightly regulated and predominantly observed in activated macrophages at the protein level, consistent with its role in innate immunity (UniProt: TISSUE SPECIFICITY).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 118-269 of mouse IL1β precursor (NP_032387.1), covering the mature IL1β region. The antibody may recognize both the ~31 kDa precursor and the ~17 kDa mature form depending on the sample and activation state. It is advisable to run appropriate molecular weight markers and positive controls (e.g., LPS-stimulated macrophage lysates) to identify the specific bands.
  • For Western blot (WB), use a reducing SDS-PAGE and standard transfer protocols. A band around 31 kDa is expected for the full-length precursor, while a band at ~17 kDa may appear in secreted or cleaved samples.
  • In immunofluorescence/ICC (IF/ICC), include stimulation controls (e.g., LPS ± ATP) to induce IL1β expression and secretion, as basal levels may be low. Co-staining with organelle markers may help visualize its cytosolic or secreted localization.
  • For ELISA, consider pairing this antibody with a suitable detection antibody recognizing a distinct epitope if used as a capture reagent. Validate the assay with recombinant mouse IL1β standards and biological samples.
  • Cross-reactivity is confirmed for mouse; perform a species-specific validation if used on other species. Endogenous mouse IgG may produce background in mouse tissues; using anti-rabbit secondary antibodies and appropriate blocking can mitigate this.
