Biological Background: IL-1β Function and Localization
- Interleukin-1 beta (IL-1β) is a potent pro-inflammatory cytokine belonging to the IL-1 family, encoded by the IL1B gene (also known as IL1F2), and is initially synthesized as an inactive precursor.
- It acts as a major endogenous pyrogen, inducing fever, prostaglandin synthesis, and broad immune activation including T-cell and B-cell proliferation and fibroblast collagen production. Related references: PMID:3920526
- IL-1β promotes Th17 cell differentiation and synergizes with IL-12 to enhance IFN-γ production from Th1 cells. Related references: PMID:10653850
- It plays a role in angiogenesis by inducing VEGF production in cooperation with TNF and IL-6. Related references: PMID:12794819
- The mature, active form of IL-1β is released from cells via gasdermin-D (GSDMD) pores during pyroptosis, linking inflammation to programmed cell death. Related references: PMID:33377178 PMID:33883744
- During Streptococcus pyogenes skin infection, IL-1β is cleaved and activated by the bacterial protease SpeB, triggering a protective inflammatory response that limits bacterial growth. Related references: PMID:28331908
- IL-1β is predominantly expressed in activated monocytes and macrophages, and localizes to the cytoplasm, lysosomes, and is secreted extracellularly, including via exosomes.
- The precursor protein has a molecular weight of approximately 31 kDa, while the mature cleaved form is about 17 kDa. The immunogen for this antibody corresponds to the mature region (amino acids 117-269).
Experimental Guidance and Technical Tips
- The immunogen covers the mature IL-1β sequence (aa 117–269), suggesting the antibody may recognize both the precursor and mature forms. In WB, consider the sample activation status and confirm detection of the relevant band(s) in your specific cell or tissue lysate.
- For IF/ICC, typical subcellular patterns include cytoplasmic and punctate lysosomal staining; appropriate fixation and permeabilization methods should be optimized for your system.
- In ELISA, ensure the antibody pair compatibility for capture/detection formats; test against recombinant mature IL-1β standards to assess binding.
- Cross-reactivity is reported for human IL-1β; validation in other species is not provided, so exercise caution when using non-human samples.
- As with any antibody, titrations and appropriate controls (positive cell lines treated with LPS, negative knockdowns) are recommended to verify specificity.
