Biological Background: IL-1 beta Function and Localization
- Interleukin-1 beta (IL-1β), also known as catabolin, is a potent pro-inflammatory cytokine initially discovered as the major endogenous pyrogen. It induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production. Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 70–170 of human IL-1β, covering the mature cytokine region. This antibody may detect both pro-IL-1β and the cleaved mature form.
- For western blotting, consider using activated monocyte or macrophage lysates (e.g., LPS-stimulated THP-1 cells) as positive controls to confirm detection of the mature ~17 kDa band and/or the ~31 kDa precursor.
- In ELISA applications, evaluating pairing with a capture antibody recognizing a non-overlapping epitope can help establish a functional sandwich assay.
- Cross-reactivity with mouse has been reported; validate detection sensitivity in the relevant murine sample system if used for mouse studies.
