Biological Background: IL-1 beta Function and Localization

  • Interleukin-1 beta (IL-1β), also known as catabolin, is a potent pro-inflammatory cytokine initially discovered as the major endogenous pyrogen. It induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production. Related references: PMID:3920526
  • Promotes Th17 differentiation of T-cells and synergizes with IL-12 to induce interferon-gamma (IFNG) synthesis from T-helper 1 (Th1) cells. Related references: PMID:10653850
  • Plays a role in angiogenesis by inducing vascular endothelial growth factor (VEGF) production synergistically with tumor necrosis factor (TNF) and interleukin-6 (IL6). Related references: PMID:12794819
  • Involved in inflammation downstream of pyroptosis: mature IL-1β is released through gasdermin-D (GSDMD) pores. Related references: PMID:33377178, PMID:33883744
  • Functions as a sensor of Streptococcus pyogenes infection in skin; it is cleaved and activated by the bacterial protease SpeB, triggering a protective inflammatory response. Related references: PMID:28331908
  • Subcellular localization includes cytoplasm, cytosol, secretory vesicles, lysosomes, and extracellular exosomes.
  • Expressed in activated monocytes/macrophages at the protein level.
  • Associated keywords: cytokine, inflammatory response, pyrogen, mitogen, secreted, lysosome.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 70–170 of human IL-1β, covering the mature cytokine region. This antibody may detect both pro-IL-1β and the cleaved mature form.
  • For western blotting, consider using activated monocyte or macrophage lysates (e.g., LPS-stimulated THP-1 cells) as positive controls to confirm detection of the mature ~17 kDa band and/or the ~31 kDa precursor.
  • In ELISA applications, evaluating pairing with a capture antibody recognizing a non-overlapping epitope can help establish a functional sandwich assay.
  • Cross-reactivity with mouse has been reported; validate detection sensitivity in the relevant murine sample system if used for mouse studies.
