Biological Background: IL1R2 Function and Localization
- IL1R2 (Interleukin-1 receptor type 2), also known as CD121b, IL1RB, and CDw121b, is a non-signaling decoy receptor for interleukin-1 cytokines, including IL1A, IL1B, and IL1RN.
- It attenuates IL1B activity by acting as a competitive binding decoy, preventing IL1B from interacting with the signaling receptor IL1R1.
- IL1R2 exists in both membrane-bound and secreted forms; the secreted form can recruit soluble IL1RAP with high affinity, forming a complex that potently neutralizes IL1B.
- The receptor preferentially binds IL1B, with distinctly weaker affinity for IL1A and IL1RN.
- IL1R2 can also modulate cellular responses through non-signaling association with IL1RAP after ligand binding.
- Subcellular localization includes the cell membrane and secretion; the protein contains immunoglobulin domains, disulfide bonds, and is glycosylated.
- The IL1R2 gene undergoes alternative splicing, producing multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 20–320) of human IL1R2, suggesting the antibody likely recognizes epitopes within the ligand-binding region; consider verifying reactivity in relevant human and mouse sample systems.
- For Western blotting, the predicted molecular weight is approximately 45 kDa; due to glycosylation, the observed band may appear slightly higher.
- ELISA applications may be suitable for detecting soluble IL1R2; validate in appropriate sample matrices and compare with established controls.
