Biological Background: IL1R1 Function and Localization
- Also known as CD121a, IL-1R-alpha, and p80, IL1R1 is the primary receptor for interleukins IL-1α, IL-1β, and the antagonist IL-1Ra. It belongs to the interleukin-1 receptor family and mediates pro-inflammatory signaling. Related references: PMID:2950091 PMID:37315560
- Upon ligand binding, IL1R1 recruits the coreceptor IL1RAP to form a high-affinity complex that activates NF-kappa-B, MAPK, and other pathways through TIR domain-mediated recruitment of adaptors TOLLIP, MYD88, and IRAK kinases.
- IL1R1 is involved in IL-1β-mediated costimulation of interferon-gamma (IFN-γ) production in T-helper 1 (Th1) cells, linking innate and adaptive immunity. Related references: PMID:10653850
- The receptor localizes to the cell membrane but can also be secreted; it contains an extracellular region with immunoglobulin-like domains and a cytoplasmic TIR domain.
- Post-translational modifications include N-linked glycosylation and disulfide bonds, and the receptor is phosphorylated upon activation.

Experimental Guidance and Technical Tips
- The immunogen corresponds to a sequence within amino acids 380–569 of human IL1R1, located in the cytoplasmic domain. Consider using a blocking peptide to confirm specificity.
- For Western blot, the predicted molecular weight is ~65 kDa for the full-length unmodified protein; however, glycosylation and other modifications may cause a shift. Validate in a relevant cell line such as Th1 cells.
- This polyclonal antibody is suitable for ELISA; optimize dilution and coating conditions for best performance. Cross-reactivity with mouse and rat IL1R1 should be verified in the respective sample systems.
