Biological Background: IL18R1/IL-18 R alpha Function and Localization
- IL18R1, also known as CD218a or IL-1Rrp, is the alpha chain of the interleukin-18 receptor and belongs to the IL-1 receptor family.
- It specifically binds the pro-inflammatory cytokine IL18 with high affinity, but does not bind IL1A or IL1B. Related references: PMID:14528293 PMID:25261253 PMID:25500532
- IL18R1 is critical for IL18-mediated signaling, leading to interferon-gamma (IFNG) synthesis from T-helper 1 (Th1) cells, a key step in inflammatory and immune responses. Related references: PMID:10653850
- The receptor is located on the cell membrane and is highly expressed in leukocytes, spleen, and lung, with lower expression in liver, small intestine, colon, prostate, thymus, placenta, and heart. It is coexpressed with IL18R1 in Th1 cells. Related references: PMID:10653850 PMID:10925275 PMID:11046021
- Post-translational modifications include disulfide bonds and glycosylation, and the protein contains immunoglobulin-like domains and transmembrane helices.
- IL18R1 contributes to IL18-induced cytokine production, functioning independently or as a complex with SLC12A3.
Experimental Guidance and Technical Tips
- Based on the immunogen design spanning the extracellular domain (amino acids 19-320), this antibody may be particularly effective in recognizing the native membrane-bound form in immunofluorescence and immunohistochemistry applications.
- For Western blot, a band around 62 kDa is expected, but additional bands may appear due to glycosylation or other modifications; consider validating with appropriate controls.
- Given the reported cross-reactivity with mouse and rat, this antibody could be used in comparative studies across these species; however, performance should be verified in the relevant biological samples.
- For ELISA, the antibody's specificity against the IL18 binding region suggests it may be suitable as a capture or detection reagent; appropriate pairing optimization is recommended.
