Biological Background: IL18 Function and Localization
- Interleukin-18 (IL-18), also known as Iboctadekin, Interferon gamma-inducing factor, and Interleukin-1 gamma, is encoded by the IL18 gene (also designated IGIF, IL1F4).
- Pro-inflammatory cytokine primarily involved in epithelial barrier repair and polarized T-helper 1 (Th1) cell and natural killer (NK) cell immune responses. Related references: PMID:10653850
- Upon binding to IL18R1 and IL18RAP, forms a signaling ternary complex which activates NF-kappa-B, triggering synthesis of inflammatory mediators. Related references: PMID:14528293 PMID:25500532 PMID:37993714
- Synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells and natural killer (NK) cells. Related references: PMID:10653850
- Involved in transduction of inflammation downstream of pyroptosis: its mature form is specifically released into the extracellular milieu through the gasdermin-D (GSDMD) pore. Related references: PMID:33883744
- Subcellular localization includes cytoplasm, cytosol, and secretion into the extracellular space.
- Expressed in ovarian carcinoma but undetectable in normal ovarian epithelial cells; resistant to proteolytic activation by caspase-1 and -4.
- Annotated with keywords: 3D-structure, Alternative splicing, Cytokine, Cytoplasm, Direct protein sequencing, Inflammatory response, Proteomics identification, Reference proteome, Secreted.
Experimental Guidance and Technical Tips
- The immunogen encompasses the full-length mature IL-18 (amino acids 1-193), which may allow detection of both precursor and cleaved forms. Consider validating the antibody’s reactivity against these forms in relevant controls.
- For WB, anticipate a band around 18 KDa corresponding to mature IL-18; a larger precursor at ~22 KDa may also be observed depending on sample preparation.
- Given the listed applications, this antibody may be suitable for multiple immunoassays. Optimize dilutions and detection conditions for each specific method (e.g., ELISA, IHC-P, IF/ICC).
- Cross-reactivity with mouse and rat enables potential use in multi-species studies. Verify performance in the species of interest using appropriate positive and negative controls.
- As IL-18 is released via GSDMD pores during pyroptosis, consider using stimuli that induce inflammasome activation when designing experiments.
