Biological Background: IL-18 Function and Localization
- Interleukin-18 (IL-18), also known as interferon-gamma-inducing factor (IGIF), Iboctadekin, or IL-1 gamma, is encoded by the IL18 gene (aliases: IGIF, IL1F4). It belongs to the IL-1 cytokine family and exists in multiple isoforms due to alternative splicing.
- IL-18 is a pro-inflammatory cytokine primarily involved in epithelial barrier repair, polarized T-helper 1 (Th1) cell and natural killer (NK) cell immune responses, and synergizes with IL-12 to induce interferon-gamma (IFN-gamma) synthesis (PubMed:10653850).
Related references: PMID:10653850 - Upon binding to its receptor complex IL18R1 and IL18RAP, IL-18 activates NF-kappa-B signaling, triggering the synthesis of various inflammatory mediators (PubMed:14528293, PubMed:25500532, PubMed:37993714).
Related references: PMID:14528293 PMID:25500532 PMID:37993714 - The mature, biologically active form of IL-18 is released into the extracellular space through gasdermin-D (GSDMD) pores during pyroptosis, linking inflammation to programmed cell death (PubMed:33883744).
Related references: PMID:33883744 - Subcellularly, IL-18 is located in the cytoplasm and cytosol, and is ultimately secreted.
- Tissue expression: IL-18 is expressed in ovarian carcinoma but is undetectable in normal ovarian epithelial cells. It is resistant to proteolytic activation by caspase-1 and -4.
- Additional keywords from UniProt: 3D-structure, alternative splicing, direct protein sequencing, and proteomics identification confirm its status as a reference proteome component.
Experimental Guidance and Technical Tips
- The immunogen (aa 37-193) corresponds to the mature IL-18 protein, targeting the active cytokine region, which should provide specificity for the biologically relevant form.
- This polyclonal antibody has been applied in WB, IP, and ELISA. For western blot, consider reducing SDS-PAGE conditions due to the predicted molecular weight of ~22 kDa.
- Given the restricted expression in ovarian carcinoma, validate the antibody in relevant human cancer cell lines or tissue samples where IL-18 is known to be upregulated.
- For ELISA and IP, optimize antibody concentrations based on your specific assay systems.
