Biological Background: IL17RD Function and Localization
- IL17RD, also known as Sef homolog (hSef) or IL17Rhom, is a transmembrane receptor belonging to the interleukin-17 receptor family. Alternative splicing produces multiple isoforms (e.g., isoform 1 and isoform 4).
- It functions as a feedback inhibitor of fibroblast growth factor (FGF)-mediated Ras-MAPK signaling, suppressing ERK activation by blocking nuclear translocation of phosphorylated ERK. Related references: PMID:12807873 PMID:12958313 PMID:15239952
- May also mediate JNK activation and apoptosis, inhibit FGF-induced FGFR1 tyrosine phosphorylation, and suppress TGFB-induced epithelial-to-mesenchymal transition.
- Localizes to the Golgi apparatus membrane, cell membrane, and cytoplasm.
- Highly expressed in vascularized tissues (kidney, colon, heart) and ductal epithelial cells; isoform 1 is abundant in fetal and adult brain, pituitary, and reproductive tissues; isoform 4 is specifically enriched in pituitary and fetal brain.
- Contains disease-associated variants linked to hypogonadotropic hypogonadism and Kallmann syndrome. The protein is glycosylated and features a signal peptide and transmembrane domain.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 150–300 of human IL17RD (NP_060033.3), a region conserved in mouse and rat, supporting the reported cross-reactivity. For Western blot, positive control lysates from kidney, colon, or heart tissues are recommended based on tissue expression data.
- The predicted molecular weight is ~82 kDa (UniProt: 82.4 kDa), but post-translational modifications such as glycosylation may cause slight migration shifts. Optimize blocking and antibody dilution for each sample type.
- For ELISA, consider using recombinant IL17RD extracellular domain fragments as capture antigen. Validate the assay with known positive/negative samples in your experimental system.
