Biological Background: IL17A Function and Localization
- Interleukin-17A (IL-17A), also known as cytotoxic T-lymphocyte-associated antigen 8 (CTLA-8), is a pro-inflammatory cytokine and member of the IL-17 family, functioning in innate and adaptive immunity (PubMed:24120361). Related references: PMID:24120361
- Signals through the IL17RA-IL17RC heterodimeric receptor complex, triggering downstream TRAF6-mediated activation of NF-kappa-B and MAPK pathways to induce expression of cytokines, chemokines, and antimicrobial peptides (PubMed:17911633, PubMed:18684971, PubMed:19825828). Related references: PMID:17911633 PMID:18684971 PMID:19825828
- As a signature effector cytokine of T-helper 17 (Th17) cells, IL-17A primarily drives neutrophil activation and recruitment to sites of infection and inflammation.
- Maintains epithelial barrier integrity by regulating tight junction biogenesis and inducing antimicrobial beta-defensins in mucosal epithelial cells (PubMed:21350122). Related references: PMID:21350122
- Involved in antiviral host defense through mechanisms such as enhancing T cell cytotoxicity and promoting B cell-mediated immune responses.
- Secreted protein expressed in memory Th17 cells, with key structural features including disulfide bonds, glycosylation, and a signal peptide.
- Keywords: Adaptive immunity, Cytokine, Disulfide bond, Glycoprotein, Immunity, Inflammatory response, Innate immunity, Secreted, Signal.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the mature IL17A protein (aa 24-155), so the antibody may recognize both the full-length secreted form and potential cleaved fragments.
- For Western blot (WB), the predicted molecular weight of IL17A is approximately 18 kDa; consider using a reducing SDS-PAGE and validating the detected band with appropriate controls.
- For ELISA, this polyclonal antibody can be used as a capture or detection reagent; consider optimizing coating concentrations and pairing with monoclonal antibodies for sandwich ELISA.
- Cross-reactivity is reported as human-specific; verify performance in relevant human samples and cell lines expressing IL17A.
- As a polyclonal antibody from rabbit, some lot-to-lot variability may be observed; always validate new batches in the intended application.
