Biological Background: IL13RA2 Function and Localization
- IL13RA2 (interleukin-13 receptor subunit alpha-2), also designated CD213A2 or IL-13-binding protein (IL-13BP), is a high-affinity receptor for interleukin-13 (IL-13).
- It functions primarily as a decoy receptor, inhibiting IL-13- and IL-4-mediated signal transduction through the JAK-STAT pathway, thereby modulating immune and inflammatory responses. Related references: PMID:11861389 PMID:17030238
- In addition to its decoy role, IL13RA2 can act as a signaling receptor in an alternative pathway involving AP-1, ultimately leading to the production of transforming growth factor beta-1 (TGFB1). Related references: PMID:16327802
- Unlike the related IL13RA1 chain, IL13RA2 lacks the cytoplasmic motifs necessary for JAK-STAT signaling, underpinning its decoy mechanism.
- The protein is localized predominantly at the cell membrane and contains a signal peptide, three extracellular fibronectin type-III repeats, a single transmembrane helix, and a short cytoplasmic tail.
- Structurally, IL13RA2 is a glycoprotein with conserved disulfide bonds, features characteristic of the type I cytokine receptor family.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 27-343), suggesting the antibody may recognize the native receptor on live cells in addition to denatured forms.
- For WB, a band at approximately 44 kDa is expected; however, glycosylation may result in a higher apparent molecular weight. Validate in the relevant sample system.
- Consider optimizing antigen retrieval and fixation conditions for IHC-P, as membrane proteins can be sensitive to processing.
- Verify cross-reactivity in mouse and rat tissues, and note that IL13RA2 expression may be upregulated under certain pathological conditions.
