Biological Background: IL13RA1 Function and Localization

  • Encoded by the IL13RA1 gene, interleukin-13 receptor subunit alpha-1 (also known as CD213A1, NR4, CT19) is a single-pass type I membrane protein and a member of the type I cytokine receptor family.
  • Contains a signal peptide, an extracellular domain with fibronectin type-III-like repeats, a transmembrane helix, and a cytoplasmic tail.
  • Functions as a low-affinity receptor for interleukin-13 (IL13); upon heterodimerization with IL4RA, forms a high-affinity functional receptor complex that mediates IL13 signaling.
  • Additionally serves as an alternate accessory protein for interleukin-4 (IL4) signaling, substituting for the common cytokine receptor gamma chain (IL2RG) but does not enhance interleukin-2 (IL2) binding.
  • Ubiquitously expressed with highest levels in heart, liver, skeletal muscle, and ovary; lower levels in brain, lung, and kidney; also detected in B-cells, T-cells, and endothelial cells (relevant to cancer/testis antigen expression).
  • Subject to post-translational modifications including N-linked glycosylation and disulfide bond formation, contributing to the observed molecular weight (~49kDa) larger than the calculated 48.8kDa.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the entire extracellular domain (amino acids 29–343) of human IL13RA1, suggesting reactivity with native and recombinant protein; consider using a positive control such as IL13RA1-overexpressing cell lysates or purified recombinant protein.
  • For Western blot, a band around 49kDa is expected; due to glycosylation, the apparent molecular weight may vary slightly; optimize blocking and antibody dilution for your system.
  • ELISA applications may benefit from pairing with a capture antibody or antigen coating; validate binding in the context of IL13 or IL4 competition if studying receptor-ligand interactions.
