Biological Background: IL12A Function and Localization
- IL12A (Interleukin-12 subunit alpha, also known as p35, CLMF, NFSK, NKSF1) is a cytokine subunit that forms the bioactive interleukin-12 (IL-12) by heterodimerizing with IL12B (p40) and interleukin-35 (IL-35) by associating with EBI3/IL27B. Related references: PMID:8605935, PMID:8943050
- IL-12 is predominantly secreted by antigen-presenting cells (B-cells, dendritic cells, macrophages, granulocytes) and critically regulates T-cell and natural killer (NK) cell responses, induces interferon-gamma (IFN-γ) production, and promotes T-helper 1 (Th1) cell differentiation, bridging innate and adaptive immunity. - This rabbit polyclonal antibody was generated against a synthetic peptide spanning amino acids 50–150 of human IL12A (NP_000873.2). It is expected to recognize both the monomeric IL12A subunit and, under non-reducing conditions, possibly the IL-12 p70 heterodimer.
- For Western blotting, a band corresponding to the 25 kDa IL12A protein is anticipated in lysates from human, mouse, and rat tissues or cell lines (e.g., activated dendritic cells, macrophages). Optimize sample preparation to retain the secreted form; include appropriate controls.
- In ELISA applications, the antibody may be used for capture or detection of native or recombinant IL-12, but careful titration and pairing with a compatible detection antibody are recommended.
- Due to its polyclonal nature, batch-to-batch consistency should be validated in the relevant experimental system prior to large-scale studies.
