Biological Background: IL-12 Subunit Beta (IL12B) Function and Localization
- Encoded by the IL12B gene, Interleukin-12 subunit beta (IL12B; also known as IL-12 p40, CLMF p40, NKSF2) is a cytokine subunit that pairs with IL12A to form the biologically active IL-12 heterodimer, and with IL23A to form IL-23.
- IL-12 acts as a growth factor for activated T cells and NK cells, enhances the lytic activity of NK/lymphokine-activated killer cells, and stimulates IFN-gamma production by resting peripheral blood mononuclear cells (PBMC).
- IL-23 plays a role in innate and adaptive immunity, and is thought to mediate acute responses to infection in peripheral tissues, while also promoting pro-inflammatory cytokine production and potentially contributing to autoimmune inflammation and tumorigenesis.
- Both IL-12 and IL-23 are secreted cytokines, suggesting that IL12B is localized to the extracellular space upon release.
- Post-translational modifications include glycosylation and disulfide bond formation, which are important for proper folding and function; the protein contains an immunoglobulin-like domain.
- The mature protein has a calculated molecular weight of approximately 37.2 kDa, consistent with the observed 37 kDa band in SDS-PAGE under reducing conditions.
Experimental Guidance and Technical Tips
- This antibody was raised against recombinant human IL12B (amino acids 23–328), encompassing the mature secreted form. It is therefore expected to recognize the properly processed cytokine.
- For Western blotting (WB), a band of approximately 37 kDa may be observed under reducing conditions in human cell lysates or tissue samples expressing IL12B. Consider using a positive control such as stimulated dendritic cells or PBMCs.
- In ELISA applications, validate the antibody’s performance in your specific assay format (e.g., direct, indirect, or sandwich ELISA) using recombinant IL12B protein as a standard.
- The antibody is reported to cross-react with human IL12B; cross-reactivity with IL12B from other species has not been evaluated and should be tested individually.
