Biological Background: Interleukin-11 Receptor Subunit Alpha Function and Localization
- IL11RA (Interleukin-11 receptor subunit alpha, also known as CRSDA) is a receptor for the cytokine IL11. It belongs to the type I cytokine receptor family and features immunoglobulin-like domains. Alternative splicing generates a soluble form (sIL11RA) that retains ligand-binding capacity.
- Membrane-bound IL11RA complexes with IL11 and the shared signal transducer IL6ST/gp130 to activate signaling pathways controlling proliferation and differentiation of skeletogenic progenitor and other mesenchymal cells. This signaling is essential for normal craniofacial bone and tooth development, restricting suture fusion and tooth number.
- The soluble form sIL11RA acts as an agonist of IL11 activity, enabling IL11 trans-signaling. The IL11:sIL11RA complex binds to IL6ST/gp130 on cells that do not express membrane-bound IL11RA, thereby expanding the cellular targets of IL11 signaling. Related references: PMID:26876177 PMID:30279168
- Subcellularly, IL11RA localizes to the membrane and can be secreted as a soluble receptor.
- IL11RA is expressed in several cell lines, including myelogenous leukemia K-562, megakaryocytic leukemia M-07e, erythroleukemia TF-1, and osteosarcoma MG-63 and SaOS-2. Expression is elevated in prostate carcinoma. Related references: PMID:7670098
- The protein is a glycoprotein with disulfide bonds and belongs to the immunoglobulin superfamily. Mutations in IL11RA are associated with craniosynostosis and other disease variants.
