Biological Background: IL10RB Function and Localization
- IL10RB (Interleukin-10 receptor subunit beta) is also known as CRFB4, IBD25, CRF2-4, D21S58, D21S66, CDW210B, IL-10R2, and IL-10RB, and is a member of the class II cytokine receptor family.
- It functions as a shared cell surface receptor required for the activation of five class 2 cytokines: IL10, IL22, IL26, IL28, and IFNL1.
- The IFNLR1/IL10RB dimer serves as a receptor for the cytokine ligands IFNL2 and IFNL3, mediating antiviral activity through stimulation of the JAK/STAT signaling pathway and induction of interferon-stimulated genes (ISGs).
- IL10RB is localized to the membrane and is a transmembrane glycoprotein containing disulfide bonds.
- It plays a role in antiviral defense and cytokine-mediated signaling, and is annotated with keywords including 3D-structure, direct protein sequencing, and proteomics identification.
- As a reference proteome member, it is a single-pass type I membrane protein with a signal peptide.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to amino acids 20-220 of human IL10RB, which encompasses the extracellular domain; this design may help in recognizing the native protein.
- In Western blot (WB), expect a band at approximately 37 kDa (predicted molecular weight: 37 kDa; UniProt: 36.995 kDa). Validate the observed band in the relevant sample system.
- For immunofluorescence (IF/ICC), membrane localization should be observed. Consider appropriate fixation and permeabilization methods to preserve epitope accessibility.
- When using ELISA, the antibody can be explored as a capture or detection reagent. Optimize coating concentrations and blocking conditions for best performance.
