Biological Background: IL10 Function and Localization

  • Interleukin-10 (IL10), also known as cytokine synthesis inhibitory factor (CSIF), is a major anti-inflammatory cytokine that acts on various immune cells to limit excessive tissue disruption caused by inflammation.
  • IL10 binds to the heterotetrameric receptor comprising IL10RA and IL10RB, leading to JAK1 and STAT2-mediated phosphorylation and activation of STAT3. Related references: PMID:16982608
  • Activated STAT3 translocates to the nucleus to drive expression of anti-inflammatory mediators and inhibit production of pro-inflammatory cytokines such as GM-CSF, G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8, and TNF in antigen-presenting cells. Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 19–178 of human IL10, covering a large portion of the mature protein, which may enable detection of multiple isoforms or precursor forms.
  • For Western blotting, expect a band near 21 kDa; however, glycosylation and other modifications may shift the apparent molecular weight. Validate using appropriate positive and negative controls.
  • In immunohistochemistry, we recommend testing known IL10-expressing tissues (e.g., inflamed or immune-rich samples) and optimizing antigen retrieval conditions.
  • For ELISA applications, this antibody can be evaluated as a capture or detection reagent, but optimal pairing conditions should be determined empirically.
