Biological Background: IL-6 Function and Localization
- IL-6 is a pleiotropic cytokine with a wide variety of biological functions in immunity, tissue regeneration, and metabolism. It binds to IL6R, then the complex associates with the signaling subunit IL6ST/gp130 to trigger intracellular IL6-signaling pathways, including classic, trans-, and cluster signaling (Probable).
- It is a potent inducer of the acute phase response. In innate immunity, it is synthesized by myeloid cells upon recognition of pathogens through toll-like receptors at sites of infection or tissue injury. In adaptive immunity, it is required for B cell differentiation into immunoglobulin-secreting cells and plays a major role in CD4+ T cell subset differentiation, such as Tfh and Th17 cells (By similarity).
- IL-6 acts as an essential factor in bone homeostasis and on vessels directly or indirectly by inducing VEGF, resulting in increased angiogenesis activity and vascular permeability. Related references: PMID:12794819 PMID:17075861
- It functions as a myokine, discharged into the bloodstream after muscle contraction, increasing lipolysis and improving insulin resistance. 'Trans-signaling' in the central nervous system also regulates energy and glucose homeostasis. Related references: PMID:20823453
- Through activation of the IL6ST-YAP-NOTCH pathway, IL-6 induces inflammation-induced epithelial regeneration. It also plays a protective role during liver injury, being required for maintenance of tissue regeneration (By similarity).
- IL-6 has a pivotal role in iron metabolism by regulating HAMP/hepcidin expression upon inflammation or bacterial infection. Related references: PMID:15124018
- The protein is secreted and subject to post‑translational modifications including glycosylation, disulfide bond formation, and phosphorylation. Keywords associated with IL-6 include acute phase, cytokine, growth factor, and reference proteome.
- Produced by skeletal muscle, IL-6 also mediates crosstalk between insulin‑sensitive tissues, intestinal L cells, and pancreatic islets to adapt to changes in insulin demand (By similarity).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full‑length mature IL‑6 protein (amino acids 1–212), suggesting the antibody may recognize both native and recombinant human IL‑6. Consider using it for detecting IL‑6 in cell culture supernatants or serum by ELISA.
- For Western blot, note the predicted molecular weight range of 21–24 kDa. Ensure appropriate sample preparation (e.g., reducing conditions) and gel percentage for resolution of low‑molecular‑weight proteins.
- As a secreted cytokine, IL‑6 may be present at low concentrations; consider sample concentration or enrichment methods for optimal detection.
- Validate antibody reactivity in your specific sample system and include appropriate positive and negative controls to confirm specificity.
