- Interleukin-36 gamma (IL-36γ) is a pro-inflammatory cytokine of the IL-1 family, also known as IL-1F9, IL-1H1, and IL-1RP2.
- It signals through the IL-36 receptor (IL1RL2/IL-36R), activating NF-κB and MAPK pathways, and shares the co-receptor IL1RAP with the IL-1 system.
- IL-36γ is predominantly expressed in epithelial barriers (skin, lung, stomach, esophagus) and drives local inflammatory responses by acting on keratinocytes, dendritic cells, and T-cells.
- In skin, it induces expression of numerous chemokines (CCL3, CCL4, CCL5, CXCL8, etc.) and pro-inflammatory mediators (TNF, S100A7, iNOS), and is upregulated in psoriatic lesions.
- In the lung, it activates fibroblasts to produce IL-8, CXCL3, and CCL20, contributing to neutrophilic airway inflammation.
- IL-36γ may play a role in innate antifungal immunity against pathogens like Aspergillus fumigatus.
- Subcellularly, IL-36γ is found in the cytoplasm and is secreted.
- The protein undergoes alternative splicing, with a predicted molecular weight of ~18.7 kDa; mature forms may be processed to ~14 kDa.
Experimental Guidance and Technical Tips
- The immunogen spans the mature protein region (aa 18-169), which may allow detection of both precursor and processed IL-36γ forms.
- For Western blot, expect bands around 14 kDa and/or 18 kDa depending on sample processing and activation state.
- In IF/ICC, use epithelial cell lines (e.g., keratinocytes) or lung fibroblasts, where IL-36γ is endogenously expressed or can be induced.
- For ELISA, testing in cell culture supernatants after stimulation (e.g., with TNF or IL-17) can enhance detection.
- The antibody is validated for human samples; cross-reactivity to other species has not been determined.
- In functional studies, consider IL-36γ upregulation in psoriatic or inflamed tissue models.
