Biological Background: IL-33R/ST2 Function and Localization
- Interleukin-1 receptor-like 1 (IL-33R/ST2), also known as Ly84, ST2, T1, is encoded by the Il1rl1 gene in mouse and is a member of the interleukin-1 receptor family.
- Acts as a receptor for interleukin-33 (IL-33), playing critical roles in innate and adaptive immunity, tissue homeostasis, and responses to environmental stresses together with coreceptor IL1RAP. Related references: PMID:17675517 PMID:18450470 PMID:22660580
- Upon IL-33 binding, recruits MYD88, IRAK1, IRAK4, and TRAF6, leading to phosphorylation of MAPK3/ERK1 and/or MAPK1/ERK2, MAPK14, and MAPK8.
- In alternatively activated macrophages, induces UCP2-dependent mitochondrial rewiring that attenuates reactive oxygen species generation and preserves the Krebs cycle for persistent itaconate production and GATA3-dependent differentiation. Related references: PMID:34644537
- Functions as an inhibitory receptor for IL-33 signaling through a splice variant.
- Subcellular localization: cell membrane (transmembrane receptor) and also secreted as a soluble form.
- Predominantly expressed in hematopoietic tissues, macrophage, erythroid, epithelial, and fibroblast cell lines; isoform A in brain astrocytes and microglia, isoform B in brain endothelial cells.
- Post-translational modifications include glycosylation, disulfide bond formation, phosphorylation, and ubiquitination; subject to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 27-332 of the extracellular domain, enabling recognition of both membrane-bound and soluble IL-33R/ST2 forms.
- For Western blot (WB), a band around 65 kDa is expected; note that glycosylation may cause a higher apparent molecular weight. Validate detection in mouse hematopoietic or macrophage cell lines (e.g., RAW 264.7).
- For immunofluorescence/immunocytochemistry (IF/ICC), confirm membrane and/or intracellular staining patterns in fixed/permeabilized cells.
- For flow cytometry (FC), use live, non-permeabilized cells to detect surface IL-33R/ST2, with appropriate isotype controls.
- For ELISA, consider pairing with a detection antibody recognizing a different epitope for sandwich assays.
- Always optimize experimental conditions (antibody dilution, incubation time, blocking) for your specific application and sample system.
