Biological Background: IL-31 Function and Localization

This recombinant protein was used as immunogen, suggesting the antibody detects the native processed form; consider using denaturing conditions for Western blot to ensure full unfolding, but verify performance in native conditions for IF/ICC.
• For Western blot, a protein of approximately 18 kDa is expected. Use a 12-15% SDS-PAGE gel for optimal resolution of low-molecular-weight proteins and transfer to PVDF or nitrocellulose membrane with 0.2 µm pore size.
• In immunofluorescence/immunocytochemistry, since IL-31 is secreted, include a protein transport inhibitor (e.g., brefeldin A) in the culture medium prior to fixation to trap intracellular protein. Validate the staining pattern with appropriate positive and negative controls.
• For ELISA, the monoclonal antibody may be evaluated as a capture or detection reagent in a sandwich format, paired with another clone or a polyclonal antibody. Test optimal coating concentrations and buffer conditions empirically.
