Biological Background: IL-27RA/WSX-1 Function and Localization
- IL-27RA/WSX-1, also known as TCCR, CRL1, or zcytor1, is the alpha subunit of the interleukin-27 receptor, a type I cytokine receptor.
- Functions as a signal-transducing subunit for the IL27/p28-EBI3 cytokine, forming a heterodimer with IL6ST/GP130 to activate STAT3 and STAT1 signaling pathways. Related references: PMID:14764690, PMID:38509369
- Also part of the humanin receptor complex, where binding of MT-RNR2/humanin triggers JAK-STAT3 signaling for neuroprotection. Related references: PMID:19386761
- Plays a role in innate defense mechanisms. Related references: PMID:14764690
- Involved in the regulation of Th1-type immune responses (By similarity).
- Localizes to the cell membrane and is highly expressed in lymphoid tissues such as spleen, lymph nodes, and peripheral blood leukocytes, with predominant expression in CD4+ T-cells.
- Structurally, it is a glycoprotein with disulfide bonds and a transmembrane helix, classified under membrane receptors.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 33–283 of the extracellular domain, suggesting the antibody targets an extracellular epitope, making it suitable for detecting native protein on cell surfaces in flow cytometry (FC).
- For ELISA applications, optimization of coating concentration and pairing with detection antibodies is recommended; consider titrating the antibody to determine the ideal working dilution.
- As a rabbit monoclonal antibody, it offers high specificity and batch-to-batch consistency; validate performance in your specific sample matrix and assay conditions.
- Due to known cross-reactivity with human and rat, this antibody can be used for comparative studies across these species; verify reactivity with appropriate positive and negative controls.
