Biological Background: IL-1RAcP Function and Localization
- IL-1RAcP (Interleukin-1 receptor accessory protein, also known as IL1R3 or C3orf13) is a coreceptor for IL-1 receptor family members, participating in IL-1, IL-33, and IL-36 signaling pathways.
- Associates with IL1R1 bound to IL1B to form the high-affinity interleukin-1 receptor complex, triggering intracellular signaling via TIR domain-mediated recruitment of TOLLIP, MYD88, and IRAK1/2, leading to NF-kB and MAPK pathway activation.
- Does not bind IL-1 independently; IL1RN (IL-1 receptor antagonist) prevents IL1R1-IL1RAcP complex formation, and the non-signaling IL1R2 decoy receptor can modulate cellular responses.
- Isoform 4 (soluble) lacks the transmembrane domain and cannot mediate canonical IL-1 signaling (PubMed:19481478); instead, it associates with secreted IL1R2 to enhance neutralization of IL1B (PubMed:12530978). Related references: PMID:19481478 PMID:12530978
- Bidirectionally involved in neuronal synapse differentiation via trans-synaptic binding to PTPRD, and may regulate NMDA receptor function by facilitating IL1B-induced Src phosphorylation.

- Given the polyclonal nature, this antibody may recognize multiple IL1RAP isoforms. Confirm detection of the specific isoform of interest in your experimental system.
- The immunogen spans the extracellular domain (aa 21-356), so the antibody is expected to detect both full-length membrane-bound and soluble secreted forms.
- For Western blotting, a band at ~65 kDa (unmodified) is expected; however, glycosylation may increase the apparent molecular weight to 70-80 kDa. Consider deglycosylation enzymes for precise size confirmation.
- In IHC-P, perform antigen retrieval and optimize primary antibody dilution; validate in known positive tissues such as liver, skin, or placenta.
- For ELISA, this antibody may serve as a capture or detection reagent; assess cross-reactivity with other IL-1 receptor family members if used in multiplex assays.
- Cross-reactivity is predicted with mouse and rat IL1RAP based on sequence homology, but confirm reactivity if moving to these species.
