Biological Background: IL-1RAcP Function and Localization
- Interleukin-1 receptor accessory protein (IL-1RAcP), also known as IL1R3 or C3orf13, is encoded by the IL1RAP gene and belongs to the interleukin-1 receptor family; it functions as a coreceptor for IL1R1, IL1RL2, and IL1RL1 in the IL-1, IL-36, and IL-33 signaling systems, respectively.
- As a coreceptor with IL1R1, IL-1RAcP forms the high-affinity interleukin-1 receptor complex upon IL-1B binding, initiating signaling cascades that involve the recruitment of adapter molecules TOLLIP, MYD88, and IRAK1/2, leading to NF-kappa-B activation.
- It does not bind interleukin-1 independently; the antagonist IL1RN prevents its association with IL1R1, and the cellular response is further modulated through non-signaling interactions with the membrane-bound decoy receptor IL1R2.
