Biological Background: IL-17RB Function and Localization

  • IL-17RB, also known as IL-17Rh1, IL-17BR, CRL4, and EVI27, is a receptor for pro-inflammatory cytokines IL-17B and IL-17E, and may play a role in controlling the growth and/or differentiation of hematopoietic cells.
  • As a transmembrane glycoprotein with a signal peptide and a single transmembrane helix, IL-17RB localizes to the cell membrane and is also secreted.
  • The IL17RB gene undergoes alternative splicing, yielding multiple transcript variants that encode different isoforms.
  • Tissue expression is predominant in endocrine organs, including fetal and adult liver, kidney, pancreas, testis, colon, brain, and small intestine, while it is not detected in peripheral blood leukocytes, lymphoid organs, and most cell lines.
  • Structural studies (3D-structure) and direct protein sequencing have contributed to the biochemical characterization of IL-17RB.
  • Keywords associated with IL-17RB include alternative splicing, glycoprotein, receptor, secreted, and transmembrane, highlighting its functional and structural attributes.

Experimental Guidance and Technical Tips

  • Given the immunogen spans amino acids 18-289 of the extracellular domain, this rabbit monoclonal antibody is expected to recognize both membrane-bound and secreted forms of human IL-17RB. For flow cytometry (FC), consider using appropriate isotype controls and optimize permeabilization conditions if intracellular isoforms are to be detected.
  • For ELISA applications, pairing with a detection antibody targeting a distinct epitope may improve assay specificity. Validate the antibody pair in a relevant human sample matrix to ensure performance.
  • Cross-reactivity is reported only with human IL-17RB; thus, validate the antibody in human-derived systems. If using model organisms, confirm homology before experimental design.
  • As a monoclonal antibody, lot-to-lot consistency is expected; however, we recommend titrating the antibody for each new application.
