Biological Background: IL-13 Function and Localization
- Interleukin-13 (IL-13) is a secreted cytokine that plays a critical role in allergic inflammation and immune responses to parasite infections. Related references: PMID:15361238
- IL-13 synergizes with IL2 to regulate interferon-gamma synthesis and stimulates the proliferation of B-cells, and the activation of eosinophils, basophils, and mast cells (By similarity).
- It downregulates proinflammatory Th1-driven responses, suppressing the synthesis of cytokines such as IL1, IL6, IL10, IL12, and TNF, partially through inhibition of NF-kappa-B (By similarity).
- IL-13 modulates the production of transmembrane and soluble forms of IL1RL1, thereby regulating IL33 activity. Related references: PMID:34789557
- Signaling is mediated via a receptor complex of IL4R and IL13RA1, which activates JAK1 and TYK2, leading to STAT6 phosphorylation. Related references: PMID:34795444, PMID:8871614
- IL13RA2 acts as a high-affinity decoy receptor that binds IL-13 and mediates its internalization and depletion. Related references: PMID:29305434
- The protein is secreted and undergoes glycosylation and disulfide bond formation.
- IL-13 induces inflammatory responses in esophageal epithelium, disrupting desmosome junctions by down-regulating DSG1. Related references: PMID:24220297
Experimental Guidance and Technical Tips
- This antibody was raised against a recombinant protein corresponding to the mature mouse IL-13 sequence (amino acids 20-131), suggesting recognition of the secreted form.
- For intracellular flow cytometry, consider using cell stimulation with PMA/ionomycin or specific antigens, followed by fixation and permeabilization to enable detection.
- For ELISA, optimize coating and detection conditions using known IL-13 standards, and validate in the relevant biological matrix.
- Since IL-13 is secreted, sample preparation should include steps to capture the cytokine in cell supernatants or serum/plasma.
