Biological Background: IKZF2 (Helios) Function and Localization
- IKZF2, also known as Helios, is a zinc finger transcription factor belonging to the Ikaros family.
- It plays a critical role in stabilizing the noninflammatory phenotype of regulatory T cells (Tregs) (By similarity).
- Helios may mediate its effects through STAT5 signaling (By similarity).
- It binds to the IL2 promoter and represses IL2 transcription via epigenetic silencing, including deacetylation of core histones (PubMed:37316189). Related references: PMID:37316189
- In Tregs, Helios is required for the recruitment of FOXP3 to the IL2 promoter (By similarity).
- IKZF2 is essential for the maturation of outer hair cells in the cochlea, and thus for normal hearing (By similarity).
- Subcellularly, IKZF2 localizes to the nucleus.
- It is expressed in various T cell types, including developing thymocytes, activated T cells, and mucosal-associated invariant T (MAIT) cells (PubMed:34826260). Related references: PMID:34826260
- IKZF2 is subject to post-translational modifications, including acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within amino acids 300-440 of human IKZF2, which may confer cross-species reactivity; verify sequence conservation if using non-human samples.
- For Western blotting, the expected molecular weight is approximately 60 kDa. Consider using lysates from Treg cells or IKZF2-expressing cell lines as positive controls.
- In ELISA, titrate the antibody against the immunogen or target protein to determine the optimal working dilution.
- As a polyclonal antibody, it may recognize multiple epitopes; validate specificity in your sample system (e.g., human, mouse, rat) with appropriate negative controls.
