Biological Background: IKKβ Function and Localization
- IKKβ, also known as IKK2 or IKBKB, is a serine/threonine kinase and the catalytic subunit of the IκB kinase (IKK) complex, essential for canonical NF-kappa-B signaling. Related references: PMID:9346484
- It phosphorylates inhibitors of NF-kappa-B (IκBs) on critical serine residues, triggering their ubiquitination and proteasomal degradation, thereby freeing NF-kappa-B to translocate to the nucleus and activate transcription of immune, proliferative, and anti-apoptotic genes. Related references: PMID:20434986, PMID:20797629, PMID:21138416
- Beyond IκBs, IKKβ phosphorylates multiple signaling components, including NEMO, RELA, NFKB1, TBK1, and IKBKE, as well as substrates such as FOXO3, NAA10, NCOA3, BCL10, IRS1, RIPK1, and IRF5, fine-tuning immune responses and cell death pathways. Related references: PMID:11297557, PMID:14673179, PMID:20410276, PMID:15084260, PMID:17213322, PMID:19716809, PMID:25326418
- Following LPS-induced TLR4 endocytosis, IKKβ phosphorylates STAT1 at Thr-749, promoting ARID5A expression and IL-6 stabilization, as well as IL-12B transcription, linking innate inflammation to adaptive immunity. Related references: PMID:38621137, PMID:32209697
- IKKβ is located in the cytoplasm, nucleus, and membrane rafts, consistent with its dynamic roles in signal transduction.
- It is highly expressed in heart, placenta, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis, and peripheral blood, reflecting its broad regulatory functions.
- Keywords highlight its involvement in phosphorylation, acetylation, ubiquitination, host-virus interactions, and disease variants, with a predicted molecular weight of ~87 kDa (though experimental detection ranges from 29–87 kDa due to isoforms/cleavage).
Experimental Guidance and Technical Tips
- For Western blot, a band corresponding to full-length IKKβ is expected near 87 kDa; however, multiple bands between 29–87 kDa may be observed due to alternative splicing, degradation, or post-translational modifications. Validate using positive controls (e.g., TNFα-stimulated HeLa or Jurkat cells) to confirm specificity.
- In ELISA, the antibody may be used as capture or detection reagent; consider pairing with a validated monoclonal antibody to IKKβ for sandwich ELISA development. Titration experiments are recommended to determine optimal working concentrations.
- Because the immunogen spans amino acids 400–669, the antibody targets the C-terminal half of the protein, which may not recognize N-terminal truncations or certain disease-associated mutants; cross-reactivity with close family members (IKKα) should be assessed experimentally using recombinant proteins or knockout lysates.
