Biological Background: IKKβ Function and Localization
- IKKβ, also known as IKK2, IKKB, or NFKBIKB, is a serine/threonine protein kinase encoded by the IKBKB gene.
- It is a central component of the canonical IκB kinase (IKK) complex, essential for NF-κB pathway activation in response to diverse stimuli such as cytokines, microbial products, and genotoxic stress. Related references: PMID:9346484
- Phosphorylates inhibitors of NF-κB (IκB) on critical serine residues, leading to their ubiquitination and proteasomal degradation, thereby releasing NF-κB for nuclear translocation and transcriptional activation of immune and survival genes. Related references: PMID:20434986 PMID:20797629 PMID:21138416
- Also phosphorylates multiple signaling substrates beyond IκB, including NEMO, RELA, NFKB1, TBK1, and IKBKE, contributing to both positive and negative feedback regulation of inflammatory signaling. Related references: PMID:11297557 PMID:14673179 PMID:20410276
- Found predominantly in the cytoplasm, but also localizes to the nucleus and membrane rafts, consistent with its roles in signal transduction and protein complex assembly.
- Highly expressed in a broad range of tissues, including heart, placenta, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis, and peripheral blood.
- Post-translationally regulated by phosphorylation, acetylation, hydroxylation, S-nitrosylation, and ubiquitination, which fine-tune its kinase activity and protein interactions.
- Genetic mutations in IKBKB are associated with severe combined immunodeficiency (SCID) and other inflammatory disorders, underscoring its clinical relevance.
Experimental Guidance and Technical Tips
- For Western blotting, the antibody detects a target at a broad molecular weight range (29-87 kDa) possibly due to isoforms or post-translational modifications; validate with appropriate positive and negative controls in your experimental system.
- In immunohistochemistry (IHC-P), antigen retrieval may be necessary; consider using citrate or EDTA-based buffers and testing in known positive tissue samples such as spleen or thymus.
- For ELISA applications, this antibody can be used for direct or indirect detection of IKKβ in cell lysates; pairing with a specific detection antibody may improve sensitivity.
