Biological Background: IKKγ Function and Localization
- IKKγ, also known as NEMO or IKBKG, is the regulatory subunit of the canonical IκB kinase (IKK) complex, essential for NF-κB signaling. It acts as a scaffold coordinating the kinase subunits IKKα and IKKβ.
- The core function of IKKγ is to facilitate the phosphorylation and subsequent ubiquitin-dependent degradation of NF-κB inhibitors (IκBs), thereby releasing NF-κB for nuclear translocation and target gene activation (PubMed:14695475, PubMed:20724660, PubMed:21518757). - It specifically recognizes and binds polyubiquitin chains, exhibiting high affinity for linear (M1-linked) and Lys-63-linked ubiquitin. - For Western blot, multiple bands may be observed (predicted molecular weights of 37, 48, and 56 kDa), possibly representing splice variants or modified protein species. Validate the detection and relative band intensity using appropriate positive and negative control lysates from human or mouse cell lines.
- In immunoprecipitation, pre-clearing steps and appropriate lysis buffer conditions should be optimized, as IKKγ exists in a large multi-protein complex.
- For ELISA applications, consider using a matched antibody pair for capture and detection, and verify sensitivity with recombinant IKKγ or known positive samples.
- Owing to the high cross-reactivity with mouse, this antibody is suitable for studies in mouse models, but validate in the specific tissue or cell type of interest.
