  • It regulates NF-κB signaling by phosphorylation of CYLD and IκB inhibitors, promoting dissociation of the NF-κB complex and subsequent nuclear activity.
  • IKKε dynamically localizes to the cytoplasm, nucleus, and PML nuclear bodies, supporting its versatile role in signal transduction.
  • The kinase also phosphorylates STAT1 at Ser-708, modulating the balance between type I (ISGF3) and type II (GAF) interferon responses.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 495–716) of human IKKε. Validate antibody specificity with positive controls (e.g., spleen lysate) and negative controls in your model system.
  • In Western blot, expect a band at ~80 kDa. Use appropriate molecular weight markers and verify the band size.
  • For IHC-P, consider antigen retrieval (e.g., citrate pH 6.0) and optimize dilution using tissues with known IKKε expression (spleen, thymus).
  • This antibody cross-reacts with mouse and rat IKKε; confirm sequence homology before application.
  • Polyclonal antibodies may exhibit lot-to-lot variability; include reference controls in each experiment.
