Biological Background: IKKα Function and Localization
- IKKα, encoded by the CHUK gene, is also known as IKK1, IKK-alpha, NFKBIKA, and TCF16; it is a conserved helix-loop-helix ubiquitous serine/threonine kinase.
- IKKα plays an essential role in NF-κB signaling, activated by inflammatory cytokines, bacterial/viral products, DNA damage, and other cellular stresses. Related references: PMID:18626576, PMID:9244310, PMID:9252186
- In the canonical pathway, it phosphorylates IκB inhibitors on serine residues, promoting their ubiquitination and proteasomal degradation, which releases NF-κB for nuclear translocation and gene transcription. Related references: PMID:18626576, PMID:35952808, PMID:9244310, PMID:9252186
- IKKα negatively regulates canonical NF-κB signaling by phosphorylating TAX1BP1, facilitating assembly of the A20/TNFAIP3 ubiquitin-editing complex to limit inflammatory gene activation. Related references: PMID:21765415
- In the non-canonical NF-κB pathway, IKKα homodimers phosphorylate NFKB2/p100, triggering its processing to p52 and activation of RelB-p52 complexes, crucial for B-cell survival and lymphoid organogenesis. Related references: PMID:20501937
- Within the nucleus, IKKα phosphorylates histone H3 at Ser10 and CREBBP, modulating chromatin accessibility and transcriptional activation at NF-κB-responsive promoters. Related references: PMID:12789342, PMID:17434128
- Additional phosphorylation targets include RIPK1 (repressing its kinase activity), AMBRA1 (promoting mitophagy), and FOXO3 (regulating apoptosis). Related references: PMID:30217973, PMID:15084260
- IKKα localizes to both cytoplasm and nucleus, with wide tissue expression, and undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 600–700) of human IKKα, ensuring specificity for the IKKα subunit over IKKβ.
- For Western blotting, expect a band at approximately 85 kDa; consider using positive and negative controls to confirm specificity.
- In immunohistochemistry and immunofluorescence, antigen retrieval and permeabilization steps should be optimized, as IKKα is present in both cytoplasmic and nuclear compartments.
- For immunoprecipitation and ELISA applications, titrate the antibody to optimal working concentrations empirically and use appropriate buffer systems.
