  • IκBα (NF-kappa-B inhibitor alpha) is a key regulator of the NF-κB signaling pathway, primarily functioning to retain NF-κB dimers in the cytoplasm.
  • It contains multiple ankyrin repeat domains that mediate protein-protein interactions, particularly with the RELA/p65 and NFKB1/p50 subunits of NF-κB.
  • Through its ankyrin repeats, IκBα masks the nuclear localization signals (NLS) of NF-κB, preventing its translocation to the nucleus.

  • Degradation of IκBα releases NF-κB, allowing it to enter the nucleus and activate transcription of target genes involved in immune and inflammatory responses.
