Biological Background: IκBα Function and Localization
- IκBα (NF-kappa-B inhibitor alpha) is a key regulator of the NF-κB signaling pathway, primarily functioning to retain NF-κB dimers in the cytoplasm.
- It contains multiple ankyrin repeat domains that mediate protein-protein interactions, particularly with the RELA/p65 and NFKB1/p50 subunits of NF-κB.
- Through its ankyrin repeats, IκBα masks the nuclear localization signals (NLS) of NF-κB, preventing its translocation to the nucleus.
- Upon stimulation by pro-inflammatory cytokines or immune signals, IκBα is phosphorylated, triggering its ubiquitination and subsequent proteasomal degradation.
- Degradation of IκBα releases NF-κB, allowing it to enter the nucleus and activate transcription of target genes involved in immune and inflammatory responses.
- IκBα shuttles between the cytoplasm and nucleus; it can also enter the nucleus to remove NF-κB from DNA and export it back to the cytoplasm.
