Biological Background: IκBα Function and Localization
- IκBα (NF-kappa-B inhibitor alpha, also known as MAD-3) is a member of the IκB family that inhibits NF-κB transcriptional activity by masking the nuclear localization signals of REL dimers (RELA/p65 and NFKB1/p50) in the cytoplasm. Related references: PMID:1493333, PMID:36651806, PMID:7479976
- Upon stimulation by immune and pro-inflammatory signals, IκBα is phosphorylated, leading to its ubiquitination and proteasomal degradation. This releases NF-κB to translocate into the nucleus and activate transcription. Related references: PMID:7479976, PMID:7628694, PMID:7796813
- IκBα shuttles between the cytoplasm and nucleus, with its nuclear export contributing to the termination of NF-κB responses.
- The protein contains ankyrin repeats and undergoes multiple post-translational modifications including phosphorylation, ubiquitination, and hydroxylation.
- Mutations in the NFKBIA gene are associated with ectodermal dysplasia and immune deficiencies.
- IκBα is also involved in host-virus interactions, as it is targeted by various viral proteins to modulate NF-κB signaling.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 201–300 (C-terminal region) of human IκBα, suggesting the antibody recognizes epitopes in this domain. Consider this when interpreting data from different species or mutants.
- For Western blotting, a band of ~36 kDa is expected based on the calculated molecular weight. Validate in relevant sample systems and use appropriate positive and negative controls.
- In immunoprecipitation, optimize the antibody concentration for efficient capture, and include controls to confirm specificity.
- For ELISA, consider pairing with a matched detection antibody if a sandwich assay is required. The polyclonal nature can provide robust signal but may require careful validation for quantitative applications.
- The antibody cross-reacts with mouse and rat IκBα, but sequence alignment of the immunogen region is recommended for other species.
