Biological Background: Ikaros Function and Localization
- DNA-binding protein Ikaros (IKZF1) is a zinc-finger transcription factor belonging to the Ikaros family, encoded by the IKZF1 gene (also known as LYF1, ZNFN1A1). It is a key regulator of hematopoietic cell differentiation (PubMed:17934067). Related references: PMID:17934067
- Ikaros binds to gamma-satellite DNA, contributing to the regulation of chromatin structure and gene expression (PubMed:17135265, PubMed:19141594). Related references: PMID:17135265 | PMID:19141594
- It plays a critical role in the development of lymphocytes, including both B and T cells, by binding and activating the enhancer of the CD3-delta gene and repressing the TdT gene during thymocyte differentiation.
- Through association with HDAC-dependent and HDAC-independent complexes, Ikaros regulates transcription, and it targets chromatin-remodeling complexes NuRD and BAF (SWI/SNF) to the beta-globin locus in adult erythrocytes via the PYR complex.
- Ikaros increases normal apoptosis in adult erythroid cells and confers early temporal competence to retinal progenitor cells (RPCs) (By similarity).
- The function of Ikaros is isoform-specific and is modulated by dominant-negative inactive isoforms (PubMed:17135265, PubMed:17934067). Related references: PMID:17135265 | PMID:17934067
- Subcellular localization is primarily nuclear, but it can also be found in the cytoplasm. Tissue expression is abundant in thymus, spleen, peripheral blood leukocytes, and lymph nodes, with lower levels in bone marrow and small intestine.
- Post-translational modifications include phosphorylation and isopeptide bond formation; the protein contains multiple C2H2-type zinc fingers essential for DNA binding.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-270) of human Ikaros, encompassing key functional domains; this design may allow recognition of multiple isoforms sharing this sequence. Validate specificity in your experimental system using appropriate controls, such as Ikaros knockdown or knockout samples.
- For Western blot (WB), consider using nuclear extracts from lymphoid tissues or cell lines (e.g., thymus, spleen, Jurkat) given the abundant expression of Ikaros in these samples. A band around 58 kDa is expected for the full-length protein.
- In immunofluorescence/immunocytochemistry (IF/ICC), Ikaros exhibits predominantly nuclear staining; ensure adequate permeabilization and consider co-staining with a nuclear marker.
- For chromatin immunoprecipitation (ChIP), verify that the antibody can efficiently immunoprecipitate Ikaros-bound genomic loci. Optimize cross-linking and sonication conditions for your cell type.
- This antibody is reported to cross-react with mouse and rat Ikaros; however, sequence alignment of the immunogen peptide across species is recommended to confirm suitability for these models.
