Biological Background: Ikaros Function and Localization

  • Ikaros (IKZF1) is a DNA-binding zinc-finger transcription factor that belongs to the Ikaros family, also known as lymphoid transcription factor LyF-1.   • It functions as a key regulator of hematopoietic cell differentiation, playing a critical role in lymphocyte development, particularly B- and T-cells.   • Ikaros modulates chromatin structure by recruiting chromatin-remodeling complexes such as NuRD and BAF (SWI/SNF) to target loci, including the beta-globin locus in adult erythrocytes, forming the PYR complex.

  • The immunogen is a synthetic peptide corresponding to amino acids 250-350 of human Ikaros, a region outside the zinc-finger domains. This may allow detection of multiple isoforms.
  • For western blotting (WB), the predicted molecular weight is approximately 58 kDa, but isoform-specific bands may appear at different sizes. Validate band patterns in relevant cell lysates (e.g., Jurkat, thymus).
  • For chromatin immunoprecipitation (ChIP) and CUT&Tag assays, due to Ikaros's association with chromatin, it is advisable to optimize fixation conditions and validate enrichment at known target genes (e.g., CD3-delta, gamma-satellite) in hematopoietic cells.
  • In immunofluorescence (IF) and immunocytochemistry (ICC), expect predominantly nuclear staining. Co-staining with a nuclear marker is recommended.
  • The antibody shows cross-reactivity with mouse samples, which may be advantageous for translational studies; however, sequence alignment of the immunogen region is suggested before use in mouse models.
