  • IHH (Indian hedgehog protein) is a member of the hedgehog family, encoded by the IHH gene. The full-length precursor undergoes autoproteolytic cleavage and cholesterol modification to generate the active N-product morphogen.
  • The N-product of IHH activates the smoothened signaling pathway by binding to patched receptors (PTCH1/2), relieving SMO inhibition, which is essential for bone and cartilage development.
  • IHH positively regulates PTHLH expression to prevent chondrocyte hypertrophy during endochondral bone formation, and independently promotes chondrocyte proliferation.

  • For Western blot, validate antibody specificity using appropriate positive controls (e.g., tissues or cells known to express IHH) and consider using a loading control. Optimize blocking and detection conditions empirically.
