Biological Background: IGHMBP2 Function and Localization
- IGHMBP2 is a probable ATP-dependent 5′-to-3′ helicase that unwinds both RNA and DNA duplexes; it has low processivity in vitro. Related references: PMID:19158098 PMID:30218034
- It specifically binds and unwinds 5′-phosphorylated single-stranded guanine-rich sequences. Related references: PMID:22999958 PMID:8349627
- It may play a role in RNA metabolism, ribosome biogenesis, or initiation of translation. Related references: PMID:19158098 PMID:19299493
- IGHMBP2 interacts with tRNA-Tyr, suggesting a function in tRNA processing or translational regulation. Related references: PMID:19299493
- The protein may also regulate transcription (by similarity).
- Subcellularly, it localizes to the nucleus, cytoplasm, and neuronal axons.
- IGHMBP2 is ubiquitously expressed with highest levels in the cerebellum, and is found in developing and adult brain.
- Mutations in IGHMBP2 are associated with neurodegenerative disorders including Charcot-Marie-Tooth disease type 2S (CMT2S) and spinal muscular atrophy with respiratory distress type 1 (SMARD1).
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 540-720 of human IGHMBP2 (NP_002171.2), a conserved region; cross-reactivity with mouse and rat is supported.
- For Western blot, a band near the predicted molecular weight of 109 kDa is expected; optimize protein loading and transfer conditions.
- For immunohistochemistry (IHC-P), antigen retrieval conditions may require optimization, and staining should be validated in tissues with known IGHMBP2 expression (e.g., cerebellum).
- Validate the antibody in the relevant sample system with appropriate controls.
