Biological Background: IGHA1 Function and Localization

IGHA1 encodes the constant region of the immunoglobulin alpha-1 heavy chain, which combines with light chains to form IgA1 antibodies. In B lymphocytes, membrane-bound IGHA1 acts as the antigen receptor; upon antigen binding, it triggers clonal expansion and differentiation into antibody-secreting plasma cells. Secreted IgA1 is the dominant antibody in mucosal secretions, mediating the effector phase of humoral immunity to eliminate bound antigens. The variable domain responsible for antigen recognition is generated by V-(D)-J rearrangement and undergoes somatic hypermutation, enabling affinity maturation.

Subcellularly, IGHA1 is localized at the plasma membrane of B cells and secreted as a soluble immunoglobulin into extracellular spaces, particularly at mucosal surfaces.

The protein contains immunoglobulin domains stabilized by disulfide bonds, is N-glycosylated, and can undergo alternative splicing and chromosomal rearrangements.

According to UniProt, the mature IGHA1 constant region has a calculated molecular weight of approximately 42.8 kDa (UniProt: 42849 Da).
Experimental Guidance and Technical Tips
- The immunogen peptide spans amino acids 122–353, covering the CH2 and CH3 domains of human IGHA1 (P01876); this may confer specificity for the IgA1 isotype over IgA2. Consider verifying subclass specificity in your experimental system.
- For Western blot (WB) under reducing conditions, a predominant band around 38 kDa may be detected, corresponding to the IGHA1 heavy chain constant region; however, the apparent molecular weight can vary due to glycosylation and association with light chains in non-reducing conditions.
- In flow cytometry (FC), intracellular staining of permeabilized B lymphocytes or IgA-secreting plasma cells may be required for optimal detection of the cytoplasmic form; surface staining of membrane IgA may also be feasible.
- As a polyclonal antibody from rabbit, it is suitable for ELISA applications; cross-reactivity with other species or immunoglobulin subclasses should be independently validated.
