Biological Background: IGFBP7 Function and Localization

  • IGFBP7 (Insulin-like growth factor-binding protein 7) is a secreted protein belonging to the IGFBP family, also designated as IGFBP-rP1, MAC25 protein, PGI2-stimulating factor, prostacyclin-stimulating factor, or tumor-derived adhesion factor (TAF).
  • Binds IGF1 and IGF2 with relatively low affinity, modulating insulin-like growth factor signaling.
  • Stimulates the production of prostacyclin (PGI2), a potent vasodilator and inhibitor of platelet aggregation (PGI2-stimulating factor activity).
  • Promotes cell adhesion, functioning as a tumor-derived adhesion factor (TAF).
  • Serves as a ligand for CD93, contributing to angiogenesis regulation.

  • Subcellularly, IGFBP7 is secreted into the extracellular space.
  • Structural features include an immunoglobulin domain, disulfide bonds, glycosylation, and phosphorylation sites.
  • The protein undergoes alternative splicing and RNA editing, generating multiple isoforms.

Experimental Guidance and Technical Tips

  • For Western blot detection, the expected molecular weight is approximately 29 kDa; note that glycosylation may result in a higher apparent molecular weight.
  • Due to the secreted nature of IGFBP7, consider enriching or using conditioned media to improve detection sensitivity.
  • In ELISA applications, the antibody may be used as a detection reagent; pairing with a capture antibody against a non-overlapping epitope is recommended for sandwich assays.
  • Cross-reacts with mouse and rat IGFBP7; validate in sample matrices relevant to your experimental model.
