Biological Background: IGF2BP3 Function and Localization
- Also known as IMP3, KOC, and VICKZ3; a member of the insulin-like growth factor 2 mRNA-binding protein family.
- Functions as an RNA-binding protein that recruits target transcripts into cytoplasmic mRNPs, shielding mRNAs from degradation and modulating translation.
- Preferentially binds to N6-methyladenosine (m6A)-modified mRNAs, enhancing their stability; for example, it binds to the coding region instability determinant (CRD) of MYC mRNA, increasing its stability in an m6A-dependent manner. Related references: PMID:29476152
- Binds to the 3'-UTR of CD44 and the 5'-UTR of IGF2, influencing cell adhesion, invadopodia formation, and growth factor regulation.
- Subcellularly localized to the nucleus, cytoplasm, P-bodies, and stress granules, reflecting its roles in mRNA transport and stress responses.
- Highly expressed during fetal development and in various cancers (e.g., cervix adenocarcinoma, pancreatic carcinoma), serving as an oncofetal protein and diagnostic marker.
- Post-translationally modified by phosphorylation and ubiquitination (isopeptide bond), with alternative splicing producing multiple isoforms.
Experimental Guidance and Technical Tips
- The antibody is raised against the N-terminal region (aa 1-200) of human IGF2BP3; consider validating specificity in your target sample system, especially if truncated isoforms are relevant.
- For Western blotting, expect a primary band near 64 kDa; additional bands may arise from modifications or isoforms. Titrate the antibody for optimal signal.
- Indicated cross-reactivity with mouse and rat allows for potential use in rodent models; always verify performance in the desired tissue or cell line.
