Biological Background: IGF2BP2/IMP2 Function and Localization
- Insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2), also known as IMP2, p62, or VICKZ family member 2, is an RNA-binding protein that plays critical roles in mRNA transport, localization, and stability.
- IGF2BP2 recruits target transcripts into cytoplasmic messenger ribonucleoprotein (mRNP) complexes, enabling mRNA transport and transient storage, and protects them from endonucleolytic cleavage and microRNA-mediated degradation.
- IGF2BP2 preferentially binds to N6-methyladenosine (m6A)-containing mRNAs and enhances their stability, contributing to post-transcriptional gene regulation. Related references: PMID:29476152
- Specifically binds to the 5'-untranslated region (5'-UTR) of IGF2 mRNAs in an isoform-specific manner, regulating expression of this growth factor. Related references: PMID:9891060

Experimental Guidance and Technical Tips
- For Western blot, the calculated molecular weight is 66 kDa; use appropriate protein markers and consider using whole cell lysates from HeLa or HEK293 cells as positive controls, as IGF2BP2 is ubiquitously expressed.
- In immunohistochemistry (IHC-P), perform antigen retrieval using heat-mediated citrate buffer (pH 6.0) and test on formalin-fixed paraffin-embedded human or mouse tissues known to express IGF2BP2, such as testis or ovary.
- For immunofluorescence/immunocytochemistry (IF/ICC), fix cells with 4% paraformaldehyde and permeabilize; validate localization patterns (cytoplasmic and nuclear) in relevant cell lines.
- The peptide immunogen lies in the C-terminal region (aa 500‑599), ensuring specificity; for ELISA, use the peptide as a positive control to confirm antibody reactivity.
- Since cross-reactivity is confirmed with mouse, this antibody can be used in murine models; however, validate in the specific strain and tissue of interest.
