Biological Background: IGF2BP2/IMP2 Function and Localization

  • Insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2, also known as IMP2, VICKZ2, and hepatocellular carcinoma autoantigen p62) is an RNA-binding protein encoded by the IGF2BP2 gene. Multiple isoforms exist due to alternative splicing and alternative initiation.
  • IGF2BP2 recruits target transcripts into cytoplasmic messenger ribonucleoprotein (mRNP) complexes, enabling mRNA transport, transient storage, and translational regulation while shielding them from endonucleolytic cleavage and microRNA-mediated degradation.
  • The protein preferentially binds N6-methyladenosine (m6A)-modified mRNAs, enhancing their stability. It stabilizes MYC mRNA by binding to its coding region instability determinant (CRD), with binding efficiency increased by m6A modification. Related references: PMID:29476152

  • For Western blotting, expect bands near 66 kDa and 62 kDa; validate with appropriate positive controls (e.g., HeLa, HepG2, or testis-derived lines).
  • In immunohistochemistry (IHC-P), optimize antigen retrieval and antibody dilution in tissues with known IGF2BP2 expression, such as testis, ovary, or certain carcinomas.
