Biological Background: IGF2BP1/IMP1 Function and Localization

  • IGF2BP1 (Insulin-like growth factor 2 mRNA-binding protein 1), also known as IMP1, ZBP1, CRD-BP, and VICKZ1, is an RNA-binding protein that recruits target transcripts into cytoplasmic mRNPs to control their transport, storage, and translation.
  • It acts as an N6-methyladenosine (m6A) reader, preferentially binding m6A-modified mRNAs and increasing their stability. This is exemplified by its interaction with the MYC mRNA coding region instability determinant (CRD), where m6A modification enhances binding, shielding the transcript from endonucleolytic cleavage and microRNA-mediated decay.   • A well-characterized function is the transport and localized translation of ACTB (β-actin) mRNA: IGF2BP1 co-transcriptionally binds the 54-nucleotide zipcode element in the ACTB 3′-UTR; the RNP is then exported and transported along the cytoskeleton to the cell periphery. Phosphorylation of IGF2BP1 at the destination releases the mRNA for local actin polymerization, a process critical for cell migration and polarity.
  • In neurons, IGF2BP1 regulates neurite outgrowth, growth cone guidance, and neuronal migration by controlling the spatial and temporal synthesis of proteins such as β-actin, and it may participate in mRNA trafficking to activated synapses.
  • The protein shuttles between the nucleus and cytoplasm and localizes to perinuclear regions, P-bodies, stress granules, and various cell projections including lamellipodia, dendrites, growth cones, filopodia, and axons.
