Biological Background: Insulin-like Growth Factor 2 (IGF2) Function and Localization
- Insulin-like growth factor 2 (IGF2), also known as IGF-II, Somatomedin-A, and T3M-11-derived growth factor, is encoded by the IGF2 gene. Alternative splicing generates multiple isoforms.
- Functions as a major fetal growth hormone, essential for fetoplacental development and tissue differentiation (By similarity). In adults, involved in glucose metabolism in adipose tissue, skeletal muscle, and liver (Probable).
- Acts as a ligand for integrin, which is required for IGF2 signaling (PubMed:28873464). Positively regulates MYOD1 transcriptional coactivator recruitment, controlling muscle terminal differentiation (By similarity). Inhibits myoblast differentiation and modulates metabolism by increasing mitochondrial respiration (By similarity). Related references: PMID:28873464
- The preptin peptide, derived from the E-domain of pro-IGF2, undergoes glucose-mediated co-secretion with insulin and amplifies glucose-mediated insulin secretion; exhibits osteogenic properties by promoting osteoblast mitogenic activity via MAPK1/3 (By similarity).

Experimental Guidance and Technical Tips
- The immunogen spans the propeptide region (aa 25–180) of human IGF2. The resulting polyclonal antibody may recognize both pro-IGF2 and mature IGF-II, as well as potential isoforms generated by alternative splicing.
- For Western blot (WB) analysis, the predicted molecular weight of the preproprotein is ~20 kDa (NP_000603.1). However, the mature secreted IGF-II peptide (~7.5 kDa) may appear as a lower band. Consider using reducing conditions and appropriate controls.
- Immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC) can benefit from tissues with high IGF2 expression, such as placenta, liver, muscle, and pancreas. Perform appropriate antigen retrieval and validate in the relevant sample system.
- ELISA applications can be developed using this antibody as a capture or detection reagent; however, optimal pairings with complementary antibodies or recombinant protein standards should be empirically determined.
