Biological Background: IGF1 Function and Localization
- Insulin-like growth factor 1 (IGF1), also known as somatomedin-C or mechano growth factor (MGF), is a secreted growth factor structurally and functionally related to insulin but with much higher growth-promoting activity. It stimulates glucose transport and glycogen synthesis in osteoblasts at concentrations lower than insulin.
- IGF1 plays a role in synapse maturation, contributing to neural development and plasticity. Related references: PMID:21076856 PMID:24132240
- Calcium-dependent exocytosis of IGF1 is required for sensory perception of smell in the olfactory bulb (by similarity).
- Acts as a high-affinity ligand for the IGF1 receptor (IGF1R), binding to its alpha subunit to activate intrinsic tyrosine kinase activity, which triggers the PI3K-AKT/PKB and Ras-MAPK signaling cascades.
- IGF1 binds to integrins ITGAV:ITGB3 and ITGA6:ITGB4, forming a ternary complex with IGFR1 essential for signal transduction. This induces phosphorylation of IGFR1, MAPK3/ERK1, MAPK1/ERK2, and AKT1. Related references: PMID:19578119 PMID:22351760 PMID:23243309
- In cardiomyocytes, IGF1 acts as a negative regulator of apoptosis via the MAPK/ERK pathway, promoting STUB1/CHIP-mediated ubiquitination and degradation of ICER-type isoforms of CREM (by similarity).
- The protein is secreted and contains three intramolecular disulfide bonds. Alternative splicing produces multiple isoforms, and mutations in the IGF1 gene are associated with deafness, dwarfism, and intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 50-150 of human IGF1, a region conserved between human and mouse, which explains the observed cross-reactivity.
- For Western blot, a band at approximately 22 kDa (mature monomer) is expected under reducing and denaturing conditions. Consider optimizing sample preparation and blocking conditions based on your experimental system.
- This antibody is suitable for ELISA and immunohistochemistry on paraffin-embedded sections (IHC-P). Antigen retrieval may be required for IHC-P, and validation in the relevant tissue or cell model is recommended.
- Given the secreted nature of IGF1, consider analyzing conditioned media or serum samples for enriched detection.
