Biological Background: IGF1 Function and Localization
- Insulin-like growth factor 1 (IGF1), also known as somatomedin-C or mechano growth factor (MGF), is a secreted protein structurally related to insulin and part of the insulin-like growth factor family.
- IGF1 exhibits potent growth-promoting activities, stimulating glucose transport, glycogen synthesis, and DNA synthesis in bone-derived osteoblastic cells, operating at lower concentrations than insulin.

- For western blotting, a band around 22 kDa is expected for the pro-form or under non-reducing conditions; due to alternative splicing and post-translational modifications, multiple bands may occur. Validate in relevant positive control lysates (e.g., human liver or osteoblast cell lines).
- For immunohistochemistry (IHC-P), antigen retrieval methods and appropriate tissue fixation should be optimized, as IGF1 is secreted and may be localized to extracellular spaces.
- In ELISA applications, pair this antibody with a matched detection antibody for sandwich ELISA formats, or use as a capture/detection antibody depending on experimental design. Verify cross-reactivity with mouse and rat samples as indicated.
